演講摘要 Our research interests are focused on understanding the biological function and biochemical mechanisms of ‘writers’ of the chromatin code. Chromatin is the complex of DNA, histones, and other proteins that constitute the physiological form of the eukaryotic genome. We study Protein Arginine Methyltransferases (PRMTs), how they catalyze arginine methylation of histone and non-histone substrates, and the biological function of arginine methylation in normal development and in cancer progression. In parallel, we study how the diverse family of histone chaperones bind and release histones and how post-translational modifications, including arginine methylation, phosphorylation, and glutamylation, regulate chaperone intrinsically disordered domains to alter histone binding. We utilize protein biochemistry and enzymology, structural biology, cancer cell culture, and eggs and embryos of the frog Xenopus laevis in our studies. These tools allow us to probe evolutionarily conserved mechanisms specifying critical events in chromatin biology. Our combined use of rigorous in vitro studies along with in vivo studies provides an uncompromised approach to fully understanding epigenetic phenomena and provide new insight into how targeting chromatin ‘writers’ could be effective chemotherapies.-Dr. David Shechter
